MOTS-c (Mitochondrial Open Reading Frame of the Twelve S rRNA type-c) is a mitochondrial-derived peptide (MDP) encoded within the 12S rRNA gene of mitochondrial DNA. This 16-amino acid peptide represents a novel class of signaling molecules that originate from the mitochondrial genome, establishing a retrograde signaling pathway from mitochondria to the nucleus. MOTS-c has been shown to regulate metabolic homeostasis by activating AMPK signaling, enhancing glucose uptake, and modulating the folate-methionine cycle. Research has demonstrated its role in improving insulin sensitivity, promoting fatty acid oxidation, and protecting against metabolic stress. The peptide has also been studied for its exercise-mimetic properties and its potential involvement in the beneficial effects of physical activity on metabolic health.
